Description

        Antiserum to human C-5 is produced in goats by immunization with purified human C-5.  The gamma globulin fraction of the absorbed, monospecific antiserum is then prepared, followed by reaction with fluorescein isothiocyanate.  The excess dye is removed by dialysis and the conjugated globulin further fractionated on DEAE cellulose to remove under- and overlabelled protein.  Total protein is determined by the Biuret reaction and the concentration of fluorescein by comparison with a fluorescein dictate standard.  Sodium azide (0.1% w/v) is added as preservative.

C-5, the fifth component of complement reacts with the C3b fragment of C3 to form C5a, and C5b.1,2.  The latter then reacts with C6 to initiate formation of the terminal active complex C5-9.  C5a, a small fragment of 8500 molecular weight, has potent anaphylotactic and chemotactic activity2.

C-5 migrates as a B1 globulin in serum and has a molecular weight of approximately 185,000.  Its average serum concentration is 7-8 mg/dl (70-80 mcg/ml).3,4
